Speakers: Emmett Sprecher (Senior Field Data Scientist, Olink Proteomics)

Original Broadcast date:  November 5, 2021

Learn from a data scientist at Olink how to get the most out of your Olink® data. The following topics will be covered in this webinar: how to use Olink® data, the statistical analysis pipeline, and combining Olink® data with other “omics” data.

Olink® data is a quantified immunoassay with qPCR or next generation sequencing (NGS) readout, and the data can be analyzed in the same manner that other expression data are analyzed. Olink® data are represented as normalized protein expression (NPX) on a log2 scale. The statistical analysis pipeline occurs in four phases, including data acquisition and cleaning, exploratory data analysis to ensure that the data look as expected, statistical analysis, and biological interpretation. Finally, a published multiomics study using Olink® technology is presented, which integrated Olink® data with mass/flow cytometry, RNA-seq, CITE-seq data in COVID-19 research. The Olink® data were validated with conventional enzyme-linked immunosorbent assay (ELISA). This study demonstrates that Olink® data complements other omics data to obtain biological insights.

The webinar covers the following points:

  • While NPX is relative, it is based on a concrete quantitative scale since an increase in 1 NPX is equivalent to ~2x increase in protein expression
  • Olink® provides some software packages for analyzing Olink® data for free
  • Olink® offers custom biostatistical services for a fee, which can include pathway enrichment analysis, extraction of known protein connections, and text mining
  • Olink® data can be integrated with other omics data for multiomics research to gain new biological insights
